Influence of protein (human galectin-3) design on aspects of lectin activity
The concept of biomedical significance of the functional pairing between tissue lectins and their glycoconjugate counterreceptors has reached the mainstream of research on the flow of biological information. A major challenge now is to identify the principles of structureâactivity relationships that underlie specificity of recognition and the ensuing post-binding processes. Toward this end, we focus on a distinct feature on the side of the lectin, i.e. its architecture to present the carbohydrate recognition domain (CRD). Working with a multifunctional human lectin, i.e. galectin-3, as model, its CRD is used in protein engineering to build variants with different modular assembly. Hereby, it becomes possible to compare activity features of the natural design, i.e. CRD attached to an N-terminal tail, with those of homo- and heterodimers and the tail-free protein. Thermodynamics of binding disaccharides proved full activity of all proteins at very similar affinity. The following glycan array testing revealed maintained preferential contact formation with N-acetyllactosamine oligomers and histo-blood group ABH epitopes irrespective of variant design. The study of carbohydrate-inhibitable binding of the test panel disclosed up to qualitative cell-type-dependent differences in sections of fixed murine epididymis and especially jejunum. By probing topological aspects of binding, the susceptibility to inhibition by a tetravalent glycocluster was markedly different for the wild-type vs the homodimeric variant proteins. The results teach the salient lesson that protein design matters: the type of CRD presentation can have a profound bearing on whether basically suited oligosaccharides, which for example tested positively in an array, will become binding partners in situ. When lectin-glycoconjugate aggregates (lattices) are formed, their structural organization will depend on this parameter. Further testing (ga)lectin variants will thus be instrumental (i) to define the full range of impact of altering protein assembly and (ii) to explain why certain types of design have been favored during the course of evolution, besides opening biomedical perspectives for potential applications of the novel galectin forms

Towards Understanding Social Sustainability: An Information Systems Research-Perspective
Although it is widely accepted to differentiate between economic, ecological, and social dimensions for sustainability, social concerns are often treated with secondary importance. In line with this, also authors from Information Systems (IS) tend, in addition to its economic obligations, to focus on the natural environment and rather neglect the social dimension or do not position their research as such. Against this backdrop, we (1) examine the status quo of IS research on social sustainability, (2) organize corresponding knowledge through a taxonomy, and (3) present specific IS-roles that foster social sustainability. Our findings help researchers and practitioners in classifying current initiatives, getting ideas for future opportunities, and moving towards an understanding of how to employ IS to achieve social goals. Moreover, this study poses a foundation for deriving relevant research questions and advanced IS-theories for social concerns. Overall, we hope to nudge discussions on this valuable pillar of sustainability

DIGITAL TRANSFORMATION IN INFORMATION SYSTEMS RESEARCH: A TAXONOMY-BASED APPROACH TO STRUCTURE THE FIELD
Digital transformation becomes increasingly important to business research and practice. Transform-ing businesses poses various opportunities (e.g., creating novel business models) but, in contrast, con-stitutes numerous of challenges (e.g., changing processes and organizational structures). As Infor-mation Systems (IS) can support such transformation, it is assumed that it has great potential in digi-talization too. Thus, there is a rising amount of digital transformation-literature in IS. However, be-cause of the popularity as well as heterogeneous fields of application and types of research, a consoli-dated overview of digital transformation is necessary. Accordingly, this study derives a taxonomy of digital transformation to structure the field. Based on (1) a literature review and (2) a taxonomy-development approach, we developed a taxonomy that consists of research-relevant aspects (e.g., most studies use qualitative approaches; limited studies use design science research) as well as topic-related aspects (e.g., manufacturing and public sectors, enterprise architecture, knowledge and tech-nology are focused). Additionally, through (3) a cluster analysis, four areas could be identified: busi-ness strategies/models, working culture, technology, and skills/knowledge. Overall, by providing such a taxonomy we aim to contribute to a better characterization of digital transformation in IS research
Elevating Social Sustainability in Business Processes: A Pattern-Based Approach
In order to face fundamental challenges of our society, sustainability has gained of increased interest for business research and practice. One approach that facilitates transforming businesses into more sustainable entities is business process management (BPM). While previous research on sustainable BPM (e.g., Green BPM) seeks to consider ecological concerns in addition to its economic obligations, social sustainability is mostly neglected in current approaches for BPM. This deficiency hinders businesses in being guided on how to systematically consider social aspects such as fair and healthy working conditions. Accordingly, this study makes use of a process pattern-based approach to provide proven, general solutions for common problems referring to social sustainability. Drawing on a literature review and expert interviews, we present a preliminary set of Socially Business Process Patterns (SBPP). Our contribution highlights best practice-solutions for practitioners and lays the foundation for more research in this field
âSORRY, TOO MUCH INFORMATIONâ DESIGNING ONLINE REVIEW SYSTEMS THAT SUPPORT INFORMATION SEARCH AND PROCESSING
Online reviews are well-known for their ability to reduce information asymmetry between sellers and buyers. However, when faced with a large number of reviews, customers can be overwhelmed by information overload. To address this problem, review systems have introduced design features intended to improve the scanning, reading and processing of online reviews. Yet, to date, we lack a comprehensive overview of these design features. Our research aims to fill this gap by consolidating the state of the art of online review system design features and develop a taxonomy for âinformation search and processing in online review systemsâ. To this end, we implement a rigorous taxonomy development process, and evaluate the resulting taxonomy by providing evidence of its completeness and usefulness. We contribute to literature and practice by introducing a new taxonomy which provides a first overview of design features of online review systems

How galectins have become multifunctional proteins
Having identified glycans of cellular
glycoconjugates as versatile molecular messages, their
recognition by sugar receptors (lectins) is a fundamental
mechanism within the flow of biological information.
This type of molecular interplay is increasingly revealed
to be involved in a wide range of (patho)physiological
processes. To do so, it is a vital prerequisite that a lectin
(and its expression) can develop more than a single skill,
that is the general ability to bind glycans. By studying
the example of vertebrate galectins as a model, a total of
five relevant characteristics is disclosed: i) access to
intra- and extracellular sites, ii) fine-tuned gene
regulation (with evidence for co-regulation of
counterreceptors) including the existence of variants due
to alternative splicing or single nucleotide
polymorphisms, iii) specificity to distinct glycans from
the glycome with different molecular meaning, iv)
binding capacity also to peptide motifs at different sites
on the protein and v) diversity of modular architecture.
They combine to endow these lectins with the capacity
to serve as multi-purpose tools. Underscoring the arising
broad-scale significance of tissue lectins, their numbers
in terms of known families and group members have
steadily grown by respective research that therefore
unveiled a well-stocked toolbox. The generation of a
network of (ga)lectins by evolutionary diversification
affords the opportunity for additive/synergistic or
antagonistic interplay in situ, an emerging aspect of
(ga)lectin functionality. It warrants close scrutiny. The
realization of the enormous potential of combinatorial
permutations using the five listed features gives further
efforts to understand the rules of functional
glycomics/lectinomics a clear direction
